Yes. That's exactly what smsflash does.

I do own a flash cart, but I have not used it during development. All
you really need is an MBV2 cable.

No problem, once your code is running you can plug it in and out all
you want. You may, however, have to reset the serial port to a sane
state before you can communicate with the PC.
